Molecular cloning and complete primary sequence of human erythrocyte porphobilinogen deaminase.
We have cloned and sequenced a cDNA clone coding for human erythrocyte porphobilinogen deaminase. It encompasses the translated region, part of the 5' and the 3' untranslated regions. The deduced 344 amino acid sequence is consistent with the molecular weight and the partial amino-acid sequence of the NH2 terminal of the purified erythrocyte enzyme. Southern analysis of human genomic DNA shows ...
متن کاملHuman adenosine deaminase. cDNA and complete primary amino acid sequence.
A previously cloned partial adenosine deaminase cDNA insert (0.8 kilobase) was used to clone additional nucleotide sequences from human HPB ALL cDNA libraries. cDNA encompassing the entire coding, and 3'-untranslated regions as well as nearly all of the 5'-untranslated region was obtained. The complete amino acid sequence of the enzyme deduced from the cDNA sequence and protein sequencing consi...
متن کاملErythrocyte porphobilinogen deaminase activity in porphyria cutanea tarda.
Porphyria cutanea tarda (PCT) results from a metabolic block in heme synthesis at the level of uroporphyrinogen decarboxylase. We measured the activity of one of the enzymes preceding it in the heme biosynthetic pathway, porphobilinogen deaminase (PBGD; EC 4.3.1.8), in erythrocytes of 47 patients with symptomatic or asymptomatic familial or sporadic PCT. PBGD activity was significantly increase...
متن کاملIsolation, Cloning and Sequence Analysis of 1-Aminocyclopropane-1-Carboxylate Deaminase Gene from Native Sinorhizobium meliloti
Background: Many plant growth-promoting bacteria including Rhizobia contain the 1-aminocyclopropane-1-carboxylate (ACC) deaminase enzyme that can leave ACC, and thereby lower the level of ethylene in stressed plants. Drought and salinity are the most common environmental stress factors for plants in Iran. Objectives: The main aim of this research was development of bio-fertilizers containing A...
متن کاملThe Mouse Porphobilinogen Deaminase Gene
The porphobilinogen deaminase gene encodes the third enzyme of the heme biosynthetic pathway. This gene is expressed in a tissue-specific manner and gives rise to two isoenzymatic forms encoded by mRNA species differing in their 5’ extremity. Recent studies in human demonstrated that the tissue-specific expression of the porphobilinogen deaminase gene is determined in erythropoietic cells, by t...
